A Silver-level 4D-structure provides a good measure of the relative occupancy of all the key conformations that the molecule naturally inhabits. Typically, the modal behaviours, preferred geometries and librational amplitudes for more than 80% of the rotatable bonds in the molecule will be well defined in a Silver structure.

We would recommend a Silver structure before undertaking any SAR analysis on a given lead compound, to inform all subsequent medicinal chemistry / lead optimisation processes.
